Expression of a resveratrol synthase gene in transgenic potato lines (Solanum tuberosum L.).

RESVERATROL is a kind of stilbenes that is found in few number of plant species, such as peanuts, mulberries and grapevines. Due to its antifungal activity and induction in response to pathogen infection, resveratrol is considered as a phytoalexin. New roles of resveratrol and/or piceid as a phytochemical of human health benefits are revealed as its association with reduced cardiovascular disease and reduced cancer risk has been demonstrated. Resveratrol production has not been examined in most transgenic plants except very few plants yet. In this study, a resveratrol synthase gene was isolated from peanut (Arachis hypogaea, L. cv. Jinpoong) plants. This gene was placed under the control of the cauliflower mosaic virus 35S promoter and introduced into two Korean varieties of potatoes (Solanum tuberosum, L. cvs. Jowon and Jasim). Putative transformants were screened by PCR with primers designed from CaMV 35S promoter and NOS terminator. Most of selected transgenic potato plants showed the amplification of 297 bp fragments by PCR of genomic DNA with RS gene specific primers, while they were absent in untransformed control plants. These plants identified to have the gene were transferred to soil to stimulate their growth. When the genomic DNAs from transgenics were digested with XbaI, the bands corresponding to the fragment containing resveratrol synthase gene was detected in all of the plants but not in the control plant. Expression of the resveratrol synthase gene was also examined by Northern blot analysis. The transformants, except one line, showed a band which was lacking in the control plant, confirming that the introduced gene is transcribed into mRNA in the transformants. The strength of the band, which reflected the level of mRNA expression, differed among the individual transformants, and a close correlation between the number of integrated genes and gene expression.
