Fine Mapping of a Quantitative Trait Locus for Resistance to Globodera pallida in tetraploid potato (Solanum tuberosum) based on population genetics.

FINE mapping of a major quantitative trait locus (QTL) for resistance to the potato cyst nematode Globodera pallida was performed using tetraploid breeding clones and varieties. Starting point was a specific PCR assay detecting haplotype ‘HC’, which was developed from Single Nucleotide Polymorphisms (SNPs) located within a resistance hot spot between marker loci GP21 and GP179 on potato chromosome V. In this region, major QTL for resistance to both late blight and G. pallida have been located. The HC marker is highly diagnostic for resistance to G. pallida pathotype Pa2/3. It is present in most varieties resistant to Globodera pallida pathotypes Pa2 and Pa3 and absent in susceptible varieties. Based on presence or absence of the HC marker, a population of tetraploid genotypes related by descent was selected for fine mapping. SNPs were identified at six loci within the interval GP21-GP179 and at two loci outside this interval on the long arm of chromosome V. The determination of linkage disequilibrium based on SNP information revealed the extent of the QRL conferring the resistance to G. pallida. In support of this research we also plan to establish the chromosomal position and physical distances of the analysed loci via Fluoresence in situ Hybridization.
