Breeding and Genetics Abdmishani, Cyrus [1], Gholami, Sara [1]. Detection and Identification of Potato Virus Y by RT-PCR and IC-RT-PCR. POTATO virus Y, a member of the largest family of viruses, Potyviridae, is capable of infecting a large range of plants in several families. It causes economically important disease and transmits through aphids and also by mechanical inoculation. Presence of this virus even in very low concentration is troublesome; therefore, rapid and accurate detecting method is highly required. RT-PCR in various modified forms can be used to differentiate viruses at the level of family, genus, species and strains. It is also fast and easy, only if, sensitive care be paid. In this study, potato samples were collected from Tehran province. Samples were tested by DAS-ELISA and inoculated to host and assay plants. Different RNA extraction methods were tested and the best one was adopted to use. RT-PCR and Immunocapture RT-PCR were carried out using general and specific primers. Products were analyzed and the accuracy of these two methods was compared to Elisa. Some times, the virus concentration is so low that Elisa tests fails to detect it but by PCR very low concentration of virus or even one particle of virus is detectable.
