Over expression of the maize Lc regulatory gene enhances anthocyanin pigmentation in specific potato cultivars. THE basic helix-loop-helix (bHLH) regulatory gene, Leaf colour (Lc), from maize is known to upregulate flavonoid biosynthesis when ectopically expressed in some Solanaceous species. Using Agrobacterium-mediated transformation we have transferred the Lc cDNA under transcriptional control of the CaMV 35S promoter into a wide range of potato cultivars. Following transformation of 28 potato cultivars, red-purple cell colonies were observed in some cultivars, primarily those with characteristic red-purple tuber pigmentation. Transformed plants were recovered in five cultivars: Purple Passion (red-purple skin, pale yellow flesh), Urenika (purple skin and purple flesh), Desiree (red skin, pale yellow flesh), Red Rascal (red skin, white flesh); and Iwa (white skin, white flesh). All transgenic lines exhibited more intense red-purple pigmentation in foliage, especially Purple Passion and Urenika. The most dramatic pigmentation change occurred in the tuber flesh. Transgenic lines of Red Rascal and Desiree both developed red-fleshed tubers, whereas Purple Passion developed purple-fleshed tubers. The purple pigmentation of tuber flesh in Urenika became more intense, whereas tuber flesh of Iwa developed a faint red pigmentation.
