Pepper - Afternoon Kim, Goon-Bo [1], Nam, Young-Woo [2], Hwang, Ju Kwang [3], Kim, Shinje [1]. Construction of 10X BAC library of Capsicum annum inbred line FP11. TWO bacterial artificial chromosome (BAC) library were constructed from hot pepper (Capsicum annum c.v. FP11) covering 10X genome. pECSBAC4 vector was used to adopt HMW-DNA digested with EcoRI or BamHI. EcoRI library and BamHI library consisted of 187 and 444 of 384 well micro-titer plates, respectively, with inserts of 120kb in average. Approximate contamination of cytoplasmic genome was estimated to be 1.1% and 1.0% for chloroplast DNA and mitochondria DNA, respectively. These libraries were used to locate CMV resistance gene cmvR1 from FP11. With a cmvR1 linked marker as a probe, 2 overlapping clones were screened out from the EcoRI library. The out-most ends of the BAC contig were used as second screening probes for the EcoRI library, however, finding any positive clone was failed. Alternatively, the BamHI library was screened to find more BAC clones containing overlapping sequences with previously screened BAC clones. As a result, a 290kb BAC contig bearing cmvR1 was constructed. This contig is being used to locate the CMV resistance gene on the pepper genome.
