Potato Genomics - Afternoon

EST-Based Analysis of Cold-Stored and Reconditioned Potato Tubers.

POTATO tubers under cold storage accumulate sugar, which causes browning of chips and French fries during frying. After cold storage, tubers usually need to be treated under warm conditions to reduce the sugar content before frying. Gene expression profile in these stored tubers is still largely unknown although there have been excellent reports at molecular biology and physiology levels on cold stored tubers. In this study, we analyzed expressed sequences tags (ESTs) of potato tubers under different temperatures, as part of the Canadian Potato Genome Project (www.cpgp.ca). Screenhouse tubers of cv Shepody were used to establish cDNA libraries for EST sequencing. The cold-stored-tuber EST library (4°C, 3 months) is a normalized library, and the reconditioned-tuber EST library (4°C for 3 months, then reconditioned at 21°C for 3 days) is a regular library. ESTs were processed, clustered, and assembled into high quality contigs. Gene ontology terms (http://www.geneontology.org/) were assigned to the sequences in the unigene sets. Approximately six thousand ESTs were generated from each library. Gene ontology annotation of ESTs and unigenes and functional annotation of library specific unigenes have been analyzed. The cold-stored-tuber library and the reconditioned-tuber library shared 565 common unigenes.
