Updates on the development of a whole genome S. habrochaites (acc. LA1777) IL population.

PLANT domestication and breeding have led to increased productivity, but, at the same time have narrowed the genetic basis of many crop species. Natural biodiversity among wild relatives of cultivated crops is a rich but under-exploited resource in plant breeding. The use of “exotic libraries”, consisting of sets of marker-defined introgression lines (ILs) originating from interspecific crosses, provides plant breeders with an efficient tool to improve the agricultural performance of modern crop varieties. A population of S. habrochaites (acc. LA1777) (H) ILs in the genetic background of S. lycopersicum (cv. E6203) (E) was developed by Monforte and Tanksley (2000). The population provides a coverage of approximately 85% of the wild genome; however, several lines still contain more than one homozygous chromosome segment introgressed from the wild species. For a more efficient use of the genetic variation harbored in this wild relative of tomato, we are developing a new set of H ILs ensuring the whole genome coverage and each carrying a single homozygous introgression from the wild species. To facilitate the development of the new ILs and sub-ILs, 150 polymorphic PCR-based markers (124 CAPS and 26 SSR) have been developed which cover the entire genome at an average distance of 10 cM. The markers have been used to genotype 104 available H ILs, and the information is being used on the derived F2 populations in order to target negative selection against the additional introgressed wild segments, and to reduce the introgressed segments’ sizes. Moreover, in order to identify wild introgressions that encompass the still uncovered regions, a BC2 population of 300 plants as well as a BC1 population of 120 plants have also been analyzed. Genotypic data of the stock lines for the 150 PCR-based markers, as well as the map position of the new sub-ILs will be presented.
