Potato Genomics - Afternoon

POCI: The design and application of a 44k oligo array as a versatile tool for the study of gene expression in potato.

WITH the ongoing development of microarray technology as well as the increasing amount of expressed gene sequence data available, the whole-transcriptome analysis for a number of crop species is within reach. Through the combined efforts of an international consortium, the construction of a 44k potato oligo array, representing the current potato unigene set, has recently been completed allowing the study of potato gene expression on a much broader level than recently thought possible. The POCI chip is designed around Agilent’s 60-mer oligo design software and SurePrint technology producing high quality and sensitivity oligo arrays. With the arrival of the POCI-array, the potential of analysing transcriptional control mechanisms in a wide range of potato research areas will be greatly enhanced. Furthermore, database exchange of expression data between consortium members will expedite the scientific progress and will provide the basis for collaborations between members of the consortium in the future. We will present data of initial validation experiments using the POCI array exploring its potential in relation to the identification of candidate genes related to potato tuber development and quality traits.
