Diagnostic procedures to assess the Distribution of Tobacco Rattle Virus in the UK.

Email: Finlay.dale@scri.ac.uk Tobacco rattle virus (TRV) causes an important blemish disease in potato tuber flesh known as spraing or corky ringspot. TRV occurs in the main areas of potato production in the UK, Europe and many temperate areas and is transmitted from plant-to-plant by free-living trichodorid nematodes. TRV has a wide host range infecting more than 400 monocotyledonous and dicotyledonous species across over 50 families. Many common agricultural weed species act as reservoirs of infection. Distribution of the virus in the soil reflects distribution of its nematode vectors. Few satisfactory methods are available for the control of TRV infection, including the use of inherent resistance and application of chemicals to control nematode movement. The field distribution of TRV/nematodes is often patchy, and the virus spreads gradually. Existing methods of detection are laborious and rely either on counting trichodorids extracted from soil samples, which may not correlate with the presence of virus, or on bait tests. In the UK bait tests can take more than a month to complete, require significant glasshouse space/labour and are expensive. Therefore, numbers of samples taken per field are usually quite small, with poor definition of affected areas, if indeed they are successfully identified. Current practice is to treat the whole field with nematicide. Recently SCRI has demonstrated that it is possible to detect the virus in the roots of many weeds within fields prior to a crop and to ‘map’ the distribution of the virus allowing growers to target risk areas for nematicide or crop avoidance. Results of detailed sampling are presented, indicated distributions of the virus within fields and possible control strategies discussed.
