Potato Genomics - Afternoon Reid, Alex [1], Hof, Lysbeth [2]. Rapid identification of potato cultivars on the European Union Common Catalogue using SSR markers. WITH the number of new potato varieties obtaining Plant Breeders’ Rights increasing yearly, the realistic and reliable maintenance of large culture collections is becoming problematic. In addition, the differentiation of varieties based on morphological characteristics is a highly skilled and time-consuming task. For these reasons, a rapid and robust method for variety identification has become extremely desirable. By screening a number of microsatellite or simple sequence repeat (SSR) markers we have identified a set of nine that can be used to differentiate all the varieties examined so far (excluding mutants of existing varieties e.g. King Edward and Red King Edward). High throughput has been obtained using multiplex reaction conditions. A database of the genetic fingerprints from several hundred varieties on the European Union Common Catalogue has been constructed against which unknown samples can be rapidly identified with great accuracy.
