Molecular analysis of the I-3 region of tomato chromosome 7.

WE are pursuing the isolation of the tomato I-3 gene for fusarium wilt resistance from Lycopersicon pennellii accession LA716 via map-based cloning. We have constructed a high-resolution map of the I-3 region on tomato chromosome 7 and have identified markers closely-flanking I-3 as well as markers co-segregating with I-3. We have also shown that I-3 is embedded in a region of microsynteny with Arabidopsis thaliana chromosome 2 and have used this microsynteny to generate new markers in the I-3 region. In addition, we have begun construction of a physical map based on these markers using BAC sequences identified by bioinformatic methods and confirmed by mapping of markers developed from BAC end sequences. We have narrowed down the location of I-3 to markers separated by approximately 50 kb in L. esculentum and are in the process of looking for resistance gene homologues that may enable identification of an I-3 gene family and ultimately lead to the identification of the I-3 gene from L. pennelli.
