Cryopreservaton linked to abiotic stress tolerance: approach for long-term conservation of potato genetic resources.

IMPROVED cryopreservation holds great potential for the long-term conservation of the Andean potato landrace collection held in-trust at CIP. Based on the reported relationships between cryopreservation and drought tolerance to dehydration resistance, we have applied cryo-methods to 4 cultivars showing different drought responses. Shoot tips, isolated from 2 and 3 week-old plantlets were exposed during 0-70 min to PVS2 cryoprotective solution. Shoot tips obtained from 3 week-old plantlets and exposed to 50 min to PVS2 showed the highest recovery rate. Shoot recovery from Desiree and Ccompis cvs was 37% and 36%, respectively. When the same treatment was applied to 1, 2, 3, and 4 week old plantlets of Desiree and Piñaza cvs., the highest average recovery rate (57.5%) ocurred with 3 week-old plantlets. The effect of cold and sucrose hardening was evaluated with 4 cultivars (Desiree, Piñaza, Ccompis and Wila Yari). Plantlets were cultured during 3 weeks under 6 and 22°C in media with 0.3M and 0.07M sucrose content. Visual observation indicated that cold reduced rooting and growth, and 0.3M sucrose reduced leaf size. Shoot tips were exposed to 50 min to PVS2. Results so far showed high survival and recovery (51%)with treatments that combines cold hardening at 6°C and media containing 0.07M sucrose. Cold hardening increased recovery and survival rates, resulting in 49% recovery, 19% over non-cold treated material (30%). Regarding sucrose hardening, 0.3M sucrose treatments resulted in lower recovery (34%) than 0.07 (45%), suggesting that tissue with high sucrose content limits shoot tips re-growth. Simultaneosly, experiment on drought tolerance is in progress. Plants were treated with 4 different water regiments. Physiological and morphological parameters are being measured, and tissue samples from each experiment collected for searching chemical cell components that correlate to cryo-ability and drought tolerance.
