Development of molecular markers for resistance to internal heat necrosis (IHN).

INTERNAL heat necrosis (IHN) is a physiological disorder of potato tubers resulting in the death and discoloration of parenchymal tissue. Environmental conditions and the large acreage of the IHN-susceptible cultivar ‘Atlantic’ result in significant losses due to the disorder in the mid-Atlantic states and Florida. As previously reported, a combination of bulk segregant analysis and AFLP marker technology was used to search for molecular markers for IHN in a small population of tetraploid 4x-2x Solanum tuberosum x S. phureja-S. stenotomum (tub x phu-stn) hybrids. These clones are being used to breed an IHN-resistant, high specific-gravity replacement for ‘Atlantic’. Five putative markers from this initial study were then tested on a much larger population of 320 clones from a combining ability study for IHN resistance/susceptibility. Two of these markers were highly significantly associated with resistance/susceptibility to IHN, but together explained only 8.6% of the variation for IHN incidence. Clearly, better markers are needed if they are to prove useful in marker-assisted selection and/or identification of candidate genes. We are currently undertaking a multifaceted approach to understanding the genetics of IHN. We will report the progress of our work in the following two areas: a repeated search for AFLP markers via bulk segregant analysis with more individuals per bulk; and QTL mapping in two distinct populations using SSR and AFLP markers.
