Physiology Shakya, Roshani [1], Navarre, Roy [2]. High-Throughput Analysis of Phenolic Metabolites in Potato Using Liquid Chromatography-Mass Spectrometry (LC-MS). A simple and fast method was developed for the extraction and simultaneous detection of vitamin C, phenolic acids, flavonoids, glycoalkaloids and anthocyanins in potato by LC-MS. More than 100 compounds were detected in 12 minute runs using a monolithic column with a DAD and MS detection. Additionally, MS/MS analysis was used to identify several unknown compounds in potato. The utility of this method is seen in that we were able to tentatively identify 36 different tuber polyamines, including kukoamines, antihypertensive compounds recently identified in potato. Polyamines have multiple roles in plants including development and disease resistance. This high throughput method can be used for “germplasm mining” to develop potatoes with enhanced nutritional properties, in identifying compounds that contribute to pest or pathogen resistance and as a tool to understand phenolic metabolism.
