Tomato sequencing - Morning

Development of tomato genome sequence infrastructure and progress toward sequencing of chromosomes 1,10 and 11.

THE US contribution to the international genome sequencing effort has centered to date on development of support infrastructure for the broader effort. Three BAC libraries (200,000 clones) have been developed and end sequenced. BAC clones have been anchored to the tomato genetic map to provide an initial resource for euchromatin sequencing with the BAC end sequence database operational for subsequent extension of minimal tiling paths. We have implemented programs for anchored BAC confirmation via both anchored marker sequencing and FISH localization and have made these tools available as a service to the members of the international effort. Continued development of the SGN database and inclusion of new tools for end sequence queries, tomato repeat sequence identification and the development of preliminary sequence annotation pipelines and visualization tools have been implemented and continue to be improved. Our sequencing efforts to date have focused on BACs near euchromatin/telomere and euchromatin/heterochromatin borders in addition to a sampling of euchromatin BACs from chromosomes 1,10 and 11. We are now turning our attention toward full euchromatin sequencing of chromosomes 1, 10 and 11 and continued expansion of the SGN database and associated sequencing support, sequence presentation and analysis tools.
