Continuation of Marker-Assisted Selection for Late Blight Resistant Potatoes.

POTATO late blight, caused by Phytophthora infestans, is one of the most damaging diseases in any crop. Deployment of resistant varieties is the most effective way to control this disease. We previously demonstrated that the high-level late blight resistance in a wild potato relative, Solanum bulbocastanum, is mainly controlled by a single resistance gene RB. Transgenic potato lines containing the RB gene have showed strong late blight resistance. This resistance is comparable to backcrossed progenies derived from somatic hybrids between potato and S. bulbocastanum. A few of these somatic hybrids are female fertile and have been used in the Wisconsin Potato Breeding Program. The progeny from these crosses cannot be easily tested for late blight resistance. We reported the development of a polymerase chain reaction-based DNA marker for tracking the RB gene in breeding populations derived from the potato-S. bulbocastanum somatic hybrids. It has been determined that the RB gene is transferred to ~30% of the plants in the next generation, which is consistent with a monogenic allele being transferred in a tetraploid state. Selection is continuing upon the BC3 populations, as well as the remaining S. bulbocastanum-related breeding lines. We are currently screening 775 plants generated from ten new breeding lines for presence of the RB gene. The future evaluation and selection of the marker-positive materials will be solely based on their agronomic performance at the 1 Hill level in 2006. Presently, sixty eight percent of these plants contain the RB gene. Sixteen marker-selected potatoes have reached the 4 Hill and 8 Hill level that were previously selected based upon agronomic traits. Continuing marker-assisted selection will facilitate faster access to environmentally friendly, RB-containing potatoes.
