TMV MP and CP transgenic co-expression alter miRNA accumulation and development in Nicotiana tabacum.

A number of different approaches have been used to identify the details of host:pathogen interactions. Recently a avenue of study of was added to the field and increasing the understanding as well as the complexity of this interaction. This new area implicates the microRNAs (miRNAs), small, endogenous RNAs that regulate gene expression in plants, in this process. Tobacco Mosaic Virus (TMV) is a member of the Tobamovirus family and encodes four major proteins: two replicase proteins, a 30 kDa movement protein (MP) and coat protein (CP). Transgenic expression of selected mutants of TMV CP confers extreme protection to infection by TMV while expression of MP enhances cell to cell movement of the virus on the infected leaf. To determine the effects of simultaneous expression of TMV MP and CP to TMV a genetic cross was made between transgenic tobacco lines that produce TMV CPT42W and MP. Unexpectedly, lines accumulating TMV MP and CPT42W presented severe abnormal phenotypes. Flower development was abnormal, leaf shape was altered, and plant height was reduced. To better define these phenotypes, we analyzed the components of the miRNA pathways as well as PTGS suppressor activity of each protein singly and in combination. Our data suggests that TMV MP interacts with TMV CP in vivo, and expression of both proteins as well as TMV infection alters accumulation of miRNAs in Nicotiana tabacum. However, no PTGS suppressor activity of MP, CP or MP/CP complex was detected. Our data suggest that viral proteins are capable of altering the miRNA pathway in absence of suppression of PTGS. The results presented in this work contribute to the understanding of the relationship between the miRNAs pathways and viral disease.
