An Integrated Cytogenetic Map of Tomato Chromosome 2. CYTOGENETIC maps incorporate data from molecular linkage maps and chromosomal features such as centromeres, telomeres and, recently, fluorescence in situ hybridization (FISH) signals. We selected 34 molecular markers obtained from RFLP that evenly cover the linkage group of tomato chromosome 2. Tomato chromosome 2 is the NOR-bearing chromosome, and corresponding to tomato linkage group 2. FISH with the bacterial artificial chromosome (BAC) of each of these DNA markers as probes was used to map their position on the meiotic pachytene chromosome. The establishment of this cytogenetic map demonstrated the different ratios of physical/genetic distance between molecular markers. We also observed that the order of the three FISH markers was reversed in relation to their order on the linkage map. Our data indicate that the deduced molecular cytogenetic map will be useful for structural and functional genomics in tomato. Log in to add this item to your schedule
