Potato Genomics - Afternoon Yakovleva, Galina [1]. Somatic fusion of potato protoplasts and primary identification of hybrid plants. WILD potato species and hybrids were used for potato useful germplasm enhancement by somatic hybridization. The mesophyll leaf protoplasts were fused in presence PEG, Ca2+, pH=10 with subsequent cultivation on media SW, CT 1, CT 2 [1]. Cultivation of fusion products includes three stages: 1) dark or diffused light, SW medium, formation of micro colonies in diameter of 1 - 1.5 mm; 2) 16 h photoperiod, 2700 lux, CT 1 medium, formation of macro colonies in diameter of 2 - 3 mm; 3) 16 h photoperiod, 2700 lux, CT 2 medium, formation of shoots of 3 – 5 mm height. The shoots were cut and passed on MS medium for rooting. The primary identification of somatic hybrids between of fusion products was conducted by peroxidase isozyme analysis of green part and roots of plants in vitro. Selected somatic hybrids were grown in vivo for two-three years (greenhouse, field) and analyzed by morphological characters. Bloomed somatic hybrids have been obtained in following combinations: 1) 78563-76cld (tbr, 4x) + S.bulbocastanum; 2) LDH (tbr, 2x) + S.bulbocastanum; 3) 78563-76cld + S.polyadenium; 4) 78563-76cld + (S.brevidens × S.etuberosum); 5) S.polyadenium + (S.etuberosum × S.brevidens); 6) S.bulbocastanum + [S.polyadenium + (S.etuberosum × S.brevidens]; 7) LDH + [S.polyadenium +(S.etuberosum × S.brevidens); 8) 86-6 (LDH × S.chacoense, 2x) + S.cardiophyllum; 9) 86-6 + S.etuberosum; 10) 86-6 + (S.etuberosum × S.brevidens).
