Exploration of carotenoid pathway regulation through combined gene expression and metabolic analysis.

RESEARCH illustrating the roles of lycopene and other carotenoids in the prevention of chronic diseases in humans has led to a need for increased understanding of the regulation of the carotenoid biosynthetic pathway and accumulation of its products in edible plant tissues. We are focusing our efforts on the model fruit system tomato as it achieves high levels of carotenoids in mature tissues and offers many genetic tools to facilitate analysis. We are taking two approaches to develop insights into regulation of carotenoid accumulation. In the first we are using S. pennellii ILs to map carotenoid QTLs and in the second we employ homozygous single, double and triple mutants in the carotenoid biosynthetic pathway to examine metabolic and gene expression regulation. Through the use of HPLC, analysis of carotenoid composition in red ripe fruits of the ILs is facilitating the identification of lines with differential carotenoid accumulation profiles for use in subsequent transcriptional profiling in the hopes of identifying candidate transcriptional regulators. The second approach combining HPLC analysis with Quantitative Real-Time PCR of carotenoid synthesis genes in the mutants is providing insights on how down regulation of some enzymes in the pathway (i.e. CrtISO in the tangerine mutant) influence expression of other pathway genes. A summary of progress via both approaches will be presented.
