Identifying and mapping resistance to green peach aphid, Myzus persicae (Sulzer), in potato.

CONTROL of Potato virus Y (PVY) and Potato leafroll virus (PLRV), the most important aphid-transmitted viruses in potato, is through seed certification to ensure virus-free seed tubers and use of pesticides to control vectors. However, vector control is of limited benefit since PVY can be acquired and spread in feeding probes of a few seconds duration and insecticides can not kill quickly enough to prevent transmission. Thus, the goal of our current potato research is to reduce virus infection and spread through host plant resistance to green peach aphid, Myzus persicae (Sulzer) and to use marker assisted selection to advance cultivar breeding. Over the past seven years, we subjected 174 advanced potato lines to intensive selection for resistance to this pest. One line that has consistently expressed resistance to both aphids was crossed with cv Chieftain to produce an F1 population. Aphids were tested for survivorship, rate of development, reproduction and adult longevity on 30 F1progeny. Life tables were constructed and intrinsic rates of increase (rm) were calculated. For green peach aphid, rm ranged from -0.046 to 0.260. We are currently screening resistant and susceptible plants using a bulked segregant analysis approach with SCAR and SSR markers which will be used to guide aphid resistance integration into advance potato breeding lines. In addition, we are using electrical penetration graphs to record feeding behavior on resistant and susceptible plants. There are no significant differences in pre-probe duration, probe duration, total time spent probing, xylem or phloem feeding (P-values: 0.252 to 0.895). With these techniques, we have ascertained that the resistance expressed in these potato lines is antixenotic.
