Use of COS markers for the Andean fruited species lulo and tree tomato.

TWO exotic species native to the Andean region (Colombia, Ecuador, and Peru), the naranjilla or lulo (Solanum quitoense) and the tamarillo or tree tomato (Solanum betaceum) have great potential to become premium products for local and export markets with a high economic return to the farmers. The Colombian Corporation for Agricultural Research (CORPOICA) maintains large collections of lulo, tree tomato, and related species. We started a collaborative project between CORPOICA and Cornell University to use COS II markers in these species for comparative genomics. 336 COS primer pairs distributed along the 12 chromosomes of tomato were amplified by PCR in different accessions of lulo and tree tomato. Of the 336 COS primer pairs, 76.5% amplified in tree tomato and 88.4% amplified in lulo. Polymorphisms (SNPs and INDELs) were found among accessions both in silico and in laboratory through CAPS markers. These results indicate that it is possible to perform comparative genomics for less developed species within the Solanaceae family through COSII sequences. It is expected that the use of these markers facilitate studies of diversity, adaptation, and a most efficient improvement of less developed solanaceae species. Currently, we are expanding the evaluation of COS II markers in different accessions of the lulo wild relative S. hirtum and the tree tomato wild relative S. uniloba. These wild relatives are able to produce fertile hybrids with lulo and tree tomato, respectively, and also bear important traits for breeding such as resistant to main biotic limitations in Colombia. Further research will be conducted to construct a comparative map for each species through the use of COSII markers, which will serve for QTL identification and marker assisted selection in breeding programs.
