Effects of quality QTLs introgression on tomato fruit proteome.

TO study the genetic control of tomato fruit quality, QTL-NILs were produced by marker assisted selection. From the cross between a cherry tomato with good overall aroma intensity and an inbred line with a common taste but with bigger fruits, five genomic regions were identified as carrying major QTLs for sugar and acid content, aroma and fruit weight and texture. The cherry tomato alleles located on chromosomes 1, 2, 4 and 9 were introgressed into the large fruit recipient genotype in order to obtain QTL-NILs. A line cumulating the five QTL regions in the recipient genotype was also obtained (named Lx). We analyzed the impact of the introgression of these regions carrying QTL on the fruit proteome. Total proteins from ripe tomato fruit pericarp were separated by two-dimensional polyacrylamide gel electrophoresis; spot intensity variations were assessed by image analysis and statistical analyses were performed on these data. Some spots showing major variations between genotypes were submitted to nano LC-MS/MS analysis for identification. Many proteins showed significant variations between the lines, demonstrating the effect of QTL introgression on the recipient line proteome. Principal component analysis of quantitatively varying spots clearly distinguished the genotypes. A correlation between the size of the introgressed regions and the number of varying spots could be pointed out. Some epistatic interactions were deduced from the comparison of the spots variations in the NILs and in Lx. The function of some identified proteins will be discussed in relation to the quality traits studied.
