Structural Homologs of CONSTANS and LEAFY in Potato and Its Wild Relatives.

AMONG the genes controlling flowering and tuberization in potato, the homologs of Arabidopsis genes CONSTANS (AtCO) and LEAFY (AtLFY) are of primary interest. Overexpression of AtCO cDNA in potato leaves impaired photoperiod-controlled tuberization (Martinez-Garcia et al., 2002). cDNA of the CONSTANS ortholog from Pharbitis nil, the species related to the Solanaceae, complemented the Arabidopsis co mutants; the transformed plants flowered much earlier than the wild type (Liu et al., 2001). In tobacco, the constitutive expression of AtLFY promoted early flowering, whereas the ectopic expression of tobacco LEAFY homolog affected only inflorescence development (Ahearn et al., 2001). Cloning tomato and potato CONSTANS homologs was reported (Ben-Naim and Lifschitz, 2003; González-Schain et al., 2003); however, only tomato sequences are presently accessible. FALSIFLORA, the AtLFY ortholog in tomato, was characterized in great detail (Molinero et al., 1999). Using the strategy based on sequence similarity, we have been cloning AtCO homologs from early and late maturing potato varieties and short-day wild Solanum species. The deduced amino acid sequences of already characterized clones from potato and S. demissum share 66% homology with Arabidopsis CONSTANS (GeneBank accession no. AAM63636) and 91% with CONSTANS 1 from S. lycopersicum (AAS67377). The nucleotide identity of AtCO homologs from potato and tomato (AY490251) is 95% for exons and 87% for introns. Partial AtLFY homologs (DQ256075, DQ256076, DQ266894, DQ266895, DQ285574-DQ285576 and DQ266893) from potato and S. demissum share 94% amino acid identity with tomato FALSIFLORA (AAF66101). Genome fragments comprising intron 2 manifest extensive infra- and intraspecific polymorphism, and the latter is not related to ploidy level among fourteen Solanum species.
